In Vitro Differentiation of Human Granulocyte / Macrophage and Erythroid
نویسنده
چکیده
The effects of recombinant human erythropoietin (Ep). granulocyte/macrophage (GM) and granulocyte (G) cobny-stimulating factors (CSF). and interleukin-3 (lL-3) on erythroid burst and GM colony growth have been studied in fetal bovine serum (FBS)-supplemented and FBS-deprived culture. Sources of progenitor cells were nonadherent or nonadherent T-lymphocyte-depleted marrow or periphoral blood cells from normal humans. G-CSF. in concentrations up to 2.3 x 10_b mob/L. induced only the formation of neutrophil colonies. In contrast. GM-CSF and 11-3 both induced GM colonies and sustained the formation of erythroid bursts in the presence of Ep. However. the activities of these growth factors were affected by the culture conditions. 11-3 induction of GM colonies depended on the presence of FBS. whereas the degree of GM-CSF induction of GM colonies in FBS-deprived cultures depended on the

Transforming Growth Factor fi Selectively Inhibits Normal and Leukemic Human Bone Marrow Cell Growth In Vitro
The effects of transforming growth factor flu or (TGF-/11 or -p2) on the in vitro proliferation and differentiation of normal and malignant human hematopoietic cells were studied. Both forms of TGF-fl suppressed both the normal cellular proliferation and colony formation induced by recombinant human interleukin-3 (IL-3) and granulocyte-macrophage colony-stimulating factor (GM-CSF). In the prese...

Effect of a New Retinoidal Benzoic Acid Derivative on Normal Human Hematopoietic Progenitor Cell Growth in Vitro1
13-cis-Retinoic acid (RA) has been demonstrated to alter hemopoiesis in vitro. We compared proliferation and differentiation effects of RA to a synthetic retinoid, (E)-4-|2-(5,6,7,8-tetrahydro-5,5,8,8-terrainethyl-2naphthalenyl-l-propenyl|benzoicacid (TTNPB), on human normal bone marrow cells. TTNPB stimulated the in vitro growth of erythroidgranulocyte-macrophage progenitors and erythroid prog...
متن کاملDistinction of human T-cell line (HUT-102)-derived activity stimulating granulocytic colony formation in diffusion chambers in vivo from activities stimulating erythroid and mixed-colony formation in vitro.
Medium conditioned in the presence of human HUT-102 T-cell line cells contains activities stimulating human mixed (colony-forming unit, erythroid, granulocyte, macrophage, megakaryocyte) and erythroid (burst-forming unit, erythroid) colony formation in methylcellulose in vitro and granulocyte colony formation in diffusion chambers in mice. The stimulatory effect of HUT-102-conditioned medium on...
متن کاملPolycythemia vera. Further in vitro studies of hematopoietic regulation.
Further in vitro studies of hematopoietic regulation were carried out in two patients with polycythemia vera who were also heterozygotes (Gd(B)/Gd(A)) for glucose-6-phosphate-dehydrogenase (G-6-PD). While only G-6-PD type A was detectable in circulating erythrocytes, granulocytes and platelets, cultures of peripheral blood and marrow from one patient revealed a substantial number of G-6-PD type...
